ABSTRACT

Background: cross-contamination between cells is a usual mistake at cell culture laboratories and cell banks worldwide. MRC5 diploid cell and Rk-13, Vero and Hela continuous cell lines are used in different stages of human viral vaccines propagation at Razi Vaccine and Serum Research Institute of Iran. However diploid and continuous cells are propagated at separated cell culture laboratory and continuous cells can contaminate MRC5 diploid cells. Therefore, a sensitive test is needed. World Health Organization recommends few molecular and cellular techniques to cell characterization


Materials and Methods: the present study was therefore designed to set up an indirect immunofluorescence [IIF] test as follows: Polyclonal anti-MRC5 cell and anti-rabbit antibodies were prepared in rabbit and goat, respectability. Anti-rabbit IgG was purified using protein G affinity chromatography, conjugated to fluorescein isothiocyanate [FITC], and then further purified to remove unbound FITC using Sephadex G 25 chromatography. Using double immunodiffusion assay, purification of homemade anti-rabbit IgG was asssayed by observation of a single arch


Results: the titer of homemade FITC conjugated goat anti rabbit IgG was measured 1/16 vs 1/8 of commercial type. Fluorescein/protein molar ratio of local made fluorescein goat anti-rabbit IgG was measured 3.44 and its protein concentration and FITC concentration were determined 1.198 mg/ml and 0.01 mg/ml, respectively


Conclusion: moreover, homemade IIF test showed 100% intra-laboratory reproducibility. Purity of three batches of MRC5 working seed cell was verified using in- house IIF test and no contamination to continuous cell lines was found

ABSTRACT

Effect of non-Objective[s]: The role of non-nutritive sucking during gavage as a nutritional intervention is well established in feeding performance and feeding skills in preterm infants. However, conflicting results on the impact of this intervention on weight gain has been reported in the infants


Method: In a double blind clinical trial, 64 preterm infants, who hospitalized in a neonatal intensive care unit [NICU], were randomized into intervention or control group [32 infants in each group]. Mothers in experimental group provided oral stimulation by pacifier during the first ten minutes of tube feeding, three times a day. Infants in control group received standard routine care. To eliminate the potentially confounding effect of maternal presence, mothers in control group were present during the gavage feeding. Infant weight without clothing was measured daily by a trained person before the morning feeding in two groups and then weigh gain were compared


Results: Despite the same caloric intake by two groups, infants in pacifier sucking group had greater daily weight gain [10.9 vs. 3.2 g, P=0.01]


Conclusion: With regards to positive effect of pacifier sucking, the application of this intervention is recommended for neonates admitted to NICU. nutritive sucking on weight gaining of preterm infants hospitalized in neonatal intensive care unit

ABSTRACT

Currently, the scientific output indexed in databases such as ISI is the major criteria for evaluation and ranking of countries, researchers, institutes and universities in the world. Each country's share in the world's scientific output in general and specifically in a specific field will be evaluated accordingly. Therefore, determination of the rate of growth of articles from Iranian authors in the ISI database in the field of immunology, its distribution and growth in different years, average citation per paper in comparison with the Iranian and world average, co-authorship, the most cited papers, the most prolific authors, the most prolific Iranian universities and institutions, are the main objectives of the study. The study area is scientometrics and the necessity of the survey methods and citation analysis has been used. Findings show that from 1974 to the end of 2010 a total of 1775 articles of Iranian writers in the immunology field have entered into ISI Web of Science. According to the results, the number of articles has been high in the recent years. Most collaboration of the Iranian authors has been with their counterparts from USA. Immunology articles have had the most interaction with the field of transplantation. Most of the articles of Iranian scholars in the field of immunology are published in the Transplantation Proceedings journal. Tehran University of Medical Sciences [TUMS] is the most active center and Ardeshir Ghavamzadeh is the most prolific author. The costs of 10.99% articles is provided by Iranian and foreign universities and institutes. Iranian papers in the field of immunology have steadily increased and in the recent years have reached its peak. Iranian immunologists have disposition to co-authorship with their counterparts from other countries which is almost the same with other subject areas in Iran. The average citation per paper in the field of immunology is higher than that of the Iranian average and lowest than the global average, much lower than global and Middle East average in the field of immunology. Tehran University of Medical Sciences is the most prolific university. Also, the most prolific authors are affiliated to the TUMS. ABSTRACT

The significant factor contributing to the distant invasion of cancer cells is the ability of tumors to produce large numbers of new blood vessels, known as angiogenesis. Many natural products inhibit angiogenesis. Herein, ethanol extract of Salvia officinalis [SO] has been analyzed for its anti-angiogenic, anti-proliferation and anti-migration activities. The anti-angiogenic effect of the SO extract was evaluated on chicken chorioallantoic membrane [CAM] neovascularisation model, microscopically. The inhibitory effect of the extract on human umbilical vein endothelial cells [HUVECs] migration was tested on the wound-healing model with an inverted microscope. In addition, SO extract was screened for its possible anti-proliferative effects by separately counting HUVECs, Wehi and K562 cells with cell counter against their control wells, So extract exhibited a significant inhibitory activity in CAM assay in a dose dependent manner. CAM angiogenesis was gradually prevented to from at 100 micro g/ml of SO extract, but completely inhibited to form at 200 micro g/ml. After human umbilical vein endothelial cells [HUVECs] were suppressed by dose-dependent SO extract, their migrations were detected by wound healing model, yet they were unable to show a dose response effect on proliferation of the different cells [50-200 micro g/ml]. As observing in this study, SOextract could inhibit proliferation of the different cells at the concentrations above 200 micro g/ml without toxic effect on the cells in doses ranged from 0-500 micro g/ml. These findings indicated that SO extract might be a promising candidate for anti-angiogenic treatment

ABSTRACT

Early separation of mother and newborn has adverse effects on various aspect of infants growth, development and breastfeeding. Regarding the long- term separation of mothers from infants in cesarean section [C/S], some strategies, such as Kangaroo mother care [KMC] is very important. So, this study was conducted to determine the effects of KMC on infant feeding pattern in hospital and exclusive breastfeeding up to six- month. In a randomized controlled trial, 160 mother-infant dyads were recruited in KMC and routine care [RC] [80 mother-infant dyads in each group]. In KMC group, 2 hours after C/S, skin to skin contact was provided with special cover. KMC was done three 60 min/day for 2 consecutive days. With taking of special cover to the KMC group and continuing this care at home, exclusive breastfeeding up to 6- month was compared between two groups. Both groups were similar regarding all background and demographic variables. In KMC group, number of feeding in hospital [p<0.05] and exclusive breastfeeding up to 6- month [p<0.05] were greater than RC, but there was no statistically difference in the time between birth and the first feeding between two groups. In KMC group, number of severe crying in hospital was lower than RC group [5.6 vs. 12.3, p<0.001]. Regarding the positive effects of KMC on breastfeeding, especially on exclusive breastfeeding up to 6- month, skin to skin contact and KMC are recommended in delivery and C/S units, and then it's continuing at home

ABSTRACT

Angiogenesis, the development of new blood vessels, is an important process in tissue development and wound healing, but becomes pathologic when associated with solid tumor growth, proliferative retinopathies, and rheumatoid arthritis. Accurate and reliable qualification of neovascular [angiogenic] response, both in vitro and in vivo, is an essential requirement for the study of new blood vessel growth. The complexity of currently used three-dimensional in vitro angiogenesis systems makes it difficult to approve material in its models. Capillary-like structure occurs on basement membrane components such as collagen and/or laminin, while in other models, CLS formation occurs on transitional matrices such as fibrin. To solve this problem, we were interested in developing an angiogenesis system which allows rapid and reliable quantification of three-dimensional neovessel formation in vitro. Human bone marrow endothelial cells were seeded on gelatin-coated microcarriers and suspended in a solution of platelet-poor plasma which was induced to polymerize by addition of calcium chloride. In this way, microcarriers were entrapped in three-dimensional coagulated plasma. Within a few hours, endothelial cells begin to leave these supporting microcarries and migrate into the coagulated-plasma matrix and formed CLS within 48-72 hours. We developed a convenient angiogenesis in vitro system which allows reliable quantification of capillary formation in a three-dimensional environment

ABSTRACT

Effects of Kangaroo mother care [KMC] has been shown for pre-term infants, however KMC has not been studied adequately in term newborns. This study was performed to evaluate the effect of Kangaroo contact [KC] on some physiological parameters in term neonates and pain score in mothers with cesarean section [C/S] in Fatemiye hospital of Shahrood [Iran]. In a randomized clinical trial, 160 mother -infant dyads divided in two KC and routine care [RC] groups. KC was done three 60 min/day for two consecutive days. After cesarean section, pain intensity was determined with verbal numeric analog sacle [0-10 score]. Temperature was observed in four sets for each neonate. Infant's reflexes scores, neonatal jaundice and urine cortisol of neonates were compared between two groups. Mother's satisfaction was recorded before discharge of hospital by checklist. Both groups were similar regarding to all baseline variables. A significant increasing in temperature was observed in half an hour [36.8 vs 36.6 [degree sign][c], P<0.05] and just before discontinuing KC [an hour after initiating KC] [36.9 vs 36.6 [degree sign][c], P<0.001]. Pain score in mother after C/S [6 vs 7.8, P<0.001], number of sever crying [5.6 vs 12.3, P<0.05] in hospital and urine cortisol before discharging were lower than RC group [388.4 vs 471.3 micro/ 24 hours, P<0.05,] There was no significant differences in jaundice and mean of reflexes score in neonates. Overall, perception of mothers was positive toward KC. This study provides evidences showing that using KC had beneficial effects for both mother and infant. In addition, in order to prevention of the complications due to separation of neonates from their mothers in C/S, skin to skin contact as a routine care is recommended in cesarean section unites in Iran

ABSTRACT

In every society, injuries and events increase along with development and progress. In Iran, death due to accidents is in second grade after death of infectious diseases. Tehran with over 10 million population and without any programming in development confronted with increase in economic and human loss at recent years. In Iran despite of physical and geographical expansion of cities and forming new event arising from elaborated societies, moreover existence of numerous sky scrapers in mega cities we don't enjoy reinforcement troops, proper equipments and facilities proportionate to accidents and events. Reinforcement at the time of accident is an important factor to reduce damages and loss at a prominent rate. Fire department has a fundamental role in reinforcement on critical circumstances. Our objective is epidemiologic examination of reported events to Tehran fire departments, human and economic damages caused by it, furthermore to reveal obstacles and problems during the time of reinforcement in every event. At this cross- sectional and observational study, all reported events to fire departments stationed has been examined in different areas of Tehran in 1379. These findings has been used in analysis. We have compared situation of events in 1379 with previous years with Chi- Suare test and descriptives schedule by using excel and spss software. Comparison of events in 1379 are trending upward in comparison with 10 years ago. The time of getting to the location 9mm, 8 7sec' on the average and obstacles were more due to traffic jam and lack of sufficient and suitable equipments for firefighters like wise narrow alley and avenue. Furthermore, injuries arising from events were more directed toward men. Distribution of events in different areas was not the same. Events in some areas were more in comparison with other areas. Most of events toke place in summer season especially on mordad and events on azar were less than other months. In the light of trending upwards of events, it is emphasized on expanding of rescue station network in different areas of Tehran and equip them improved reinforcement equipment, increase in skilled human resources, moreover public training of citizens
